The family consists of four roundabout (Robo) receptors (ROBO1-4) that play a role in the development of the central nervous system during neuronal axon guidance. ROBOs is a part of the larger Ig-superfamily and consists of a series of Fibronectin-type 3 domains followed by a series of Ig-like domains that are involved in cis/trans interactions across the developing and established synapses. They serve as receptors for multiple ligands including Slits, a group of extracellular matrix proteins secreted by the midline of the nervous system that function as repellents in axonal growth. 1 2
